Reconstitution Guide
Lyophilized (freeze-dried) peptides arrive as a powder cake. Reconstitution is the process of returning them to solution for dosing — and it is where most handling errors happen.
What you need
- Lyophilized peptide vial (peptide powder cake)
- Solvent of choice — bacteriostatic water (BAC) for multi-use, sterile water for immediate single use (see BAC vs Sterile Water)
- Alcohol prep pads, 1 mL insulin syringes, and a clean workspace
Step-by-step
- Sanitize. Wipe the vial stopper with an alcohol pad and let it dry. Wash hands.
- Draw solvent. Draw the desired volume of BAC into a syringe (see the calculator for volumes).
- Inject slowly. Insert the needle through the center of the stopper at a slight angle. Inject the solvent slowly, down the inside wall of the vial — never directly onto the peptide cake.
- Roll, don't shake. Gently swirl or roll the vial. Shaking creates bubbles and shear stress that can degrade fragile peptides.
- Let it settle. If the cake hasn't fully dissolved, let the vial sit for a few minutes. Some peptides (e.g. GHK-Cu) dissolve slower or leave a slight hue — that's normal.
- Refrigerate. Store reconstituted peptide in the fridge. See Storage & Handling for timelines.
Common mistakes
Injecting solvent directly onto the peptide cake. This is the #1 cause of degradation complaints — it can denature the peptide before it ever leaves the vial.
- Over-diluting: huge volumes make doses awkwardly large. 1–2 mL per 5 mg vial is typical.
- Under-diluting: 0.5 mL into a 10 mg vial gives a concentration that is hard to dose precisely and can be painful to inject.
- Using the wrong solvent: sterile water has no preservative; anything drawn from that vial after the first puncture is a contamination risk.
- Skipping alcohol prep: the stopper is not sterile. Wipe it.
Solvent choice in one line
Multi-dose vial, dosing over days/weeks → bacteriostatic water. Single preparation, immediate use → sterile water is acceptable. Anything else is a trade-off you should make consciously.
