Adipotide
Investigational prohibitin-targeting pro-apoptotic peptide conjugate studied in obese mice and monkeys to reduce adipose vasculature; not tested in humans.
Overview
Adipotide is an investigational peptide conjugate developed for the targeted reduction of white adipose tissue. It combines a short homing peptide that binds prohibitin, a protein exposed on the surface of adipose-tissue vasculature, with a pro-apoptotic peptide sequence intended to kill the targeted endothelial cells.
Research was conducted in obese mouse models and in obese rhesus monkeys by an academic group; the strategy was to destroy the blood supply of fat tissue so that it is reabsorbed. The approach did not advance into human trials.
Mechanism
By targeting prohibitin on the endothelial cells of adipose vasculature, the conjugate is designed to deliver a pro-apoptotic (KLAKLAK-type) peptide into those cells, triggering cell death and reducing the vascular bed that supports fat tissue.
Dosing
All available data come from animal studies; no human dosing exists. In primate work the conjugate was administered by intravenous injection on a repeated schedule over several weeks.

Expected Effects

Side Effects & Tolerability
In the non-human primate study, kidney-related laboratory changes were reported in some animals, raising safety concerns for further development. Human tolerability has not been studied.
