Overview
This entry covers the research-use combination of CJC-1295 and ipamorelin, two synthetic peptides acting on different points of the growth hormone (GH) axis. CJC-1295 is a long-acting analog of growth-hormone-releasing hormone (GHRH), developed with drug-affinity-complex (DAC) technology intended to prolong its action; ipamorelin is a synthetic ghrelin-mimetic growth hormone secretagogue with selectivity for the GH secretagogue receptor.
The combination is used in research and off-label contexts on the rationale that stimulating both GHRH- and ghrelin-type pathways produces more robust GH pulses. Published human data on the combination itself are scarce, although the individual components have reached early clinical testing in other contexts.
Mechanism
CJC-1295 activates the GHRH receptor on pituitary somatotrophs, increasing the amplitude of GH pulses. Ipamorelin mimics ghrelin's action at the GH secretagogue receptor, augmenting GH release through a complementary pathway.
Note that some vendors supply CJC-1295 without the DAC modification; that form behaves more like the shorter-acting GHRH(1-29) peptide (Mod GRF 1-29) and should not be assumed equivalent to the DAC-conjugated version.
Dosing
No approved or validated dosing exists. In research-vendor use, both peptides are given by subcutaneous injection, with protocols varying widely.
