Overview

Delta sleep-inducing peptide (DSIP) is a nonapeptide first described in the late 1970s, following experiments in which a sleep-promoting factor was isolated from the blood of rabbits induced to sleep. Its name reflects early observations that it increased delta (slow-wave) sleep in animal recipients.

Despite decades of research interest, DSIP's role in human sleep regulation remains uncertain, and published human studies have produced inconsistent results. It persists as a commonly listed research peptide with proposed actions beyond sleep.

Mechanism

No clearly defined receptor or signaling pathway for DSIP has been established. Proposed mechanisms have included modulation of central neurotransmitter systems, effects on the hypothalamic-pituitary-adrenal axis, and influences on neuropeptide processing, but the evidence base is old and limited.

Reported effects in animal work include alterations in sleep architecture, stress-hormone patterns, pain sensitivity, and some metabolic parameters; independent replication has been inconsistent.

Dosing

No standard or validated dosing exists. In animal studies the peptide is often given centrally (intracerebroventricularly) or systemically at microgram-scale amounts per animal; early human studies used intravenous administration before sleep.
