FOXO4-DRI
Synthetic D-retro-inverso peptide designed to disrupt FOXO4-p53 signaling and selectively clear senescent cells in preclinical models.
Overview
FOXO4-DRI is an experimental synthetic peptide built from the FOXO4 interaction domain in D-retro-inverso form, meaning it consists of D-amino acids arranged in a reversed sequence. The design is intended to resist enzymatic breakdown and to enter cells, where the peptide can competitively interfere with binding between the transcription factor FOXO4 and p53.
It entered the senescence field through a widely cited 2017 study (Baar et al., Cell) reporting that the peptide induced apoptosis of senescent cells in culture and in aged or chemotherapy-treated mice. Published work to date remains preclinical.
Mechanism
Senescent cells resist apoptosis partly because FOXO4 sequesters p53 away from the nucleus. FOXO4-DRI mimics part of the FOXO4-p53 interface; by displacing p53 it is proposed to restore p53 activity and push senescent cells into cell death while leaving proliferating cells largely untouched. Clearing senescent cells is also expected to reduce the senescence-associated secretory phenotype, the mix of inflammatory factors these cells release.
Dosing
FOXO4-DRI has been used only in laboratory and animal research. In published mouse studies it is delivered by injection at protocol-specific doses, and no human research dosing has been established. Dosing details should be taken from the individual publications.
Expected Effects
- reduced senescent-cell burden in treated tissues (cell and rodent studies)
- improved aging- and chemotherapy-related tissue dysfunction reported in the 2017 mouse study, including hair regrowth and markers of kidney and skin health
- lower levels of senescence-associated secretory phenotype markers
- restored proliferative cell markers in treated tissue
Side Effects & Tolerability
Tolerability data are limited to rodents and cell models; the published animal work did not report major treatment-related toxicity, but systematic toxicology, biodistribution, and long-term safety datasets are lacking. Because p53 is central to tumor suppression, long-term consequences of forcing senescent-cell apoptosis remain an open research question.
Storage
As a lyophilized synthetic peptide, FOXO4-DRI should be stored desiccated, protected from light, and kept frozen at -20 deg C or below. After reconstitution in a suitable sterile solvent, solutions are generally stable for only a short period refrigerated at 2-8 deg C, and repeated freeze-thaw cycles should be avoided.
