Mazdutide
Dual GLP-1/glucagon receptor agonist peptide in clinical development for obesity and type 2 diabetes.
Overview
Mazdutide is an investigational synthetic peptide that acts as an agonist at both the glucagon-like peptide-1 (GLP-1) receptor and the glucagon receptor. It is being developed primarily in China by Innovent Biologics, with chemistry licensed from Eli Lilly, as a once-weekly treatment for type 2 diabetes and obesity. Published phase 2 results in Chinese adults with overweight, obesity, or type 2 diabetes reported dose-dependent weight loss and glycemic improvements.
Mechanism
Dual agonism is designed to combine the appetite-suppressing and insulinotropic effects of GLP-1 with the energy-expenditure and lipolytic effects of glucagon receptor signaling. The balance of the two activities is intended to produce greater weight loss than GLP-1 agonism alone while minimizing glucagon-driven hyperglycemia.
Dosing
In clinical trials mazdutide is administered as a once-weekly subcutaneous injection at dose levels defined by each study protocol. It is an investigational drug; dosing outside registered trials is not appropriate.
Expected Effects
- dose-dependent body-weight reduction in phase 2 trials in adults with overweight or obesity
- reductions in HbA1c in trials enrolling people with type 2 diabetes
- improvements in some cardiometabolic markers in early studies
Side Effects & Tolerability
Reported adverse events are typical of the incretin class, principally gastrointestinal (nausea, vomiting, diarrhea), with severity increasing at higher doses. Long-term safety data from larger phase 3 programs are still being generated.
