PNC-27

A chimeric p53-derived research peptide studied preclinically for selective membrane-mediated killing of cancer cells.

Overview

PNC-27 is a chimeric research peptide that links the HDM2-binding region of the tumor-suppressor protein p53 to a membrane-penetrating (cell-penetrating) sequence. It is studied as a potential anticancer agent, with work reported mainly in cancer cell lines and tumor-bearing animal models. Human data do not exist.

Mechanism

The proposed mechanism is unusual: rather than relying on p53 transcriptional activity, PNC-27 is reported to insert into and permeabilize the plasma membrane of cancer cells, causing rapid cell death, while normal cells are relatively spared. The selectivity is attributed to differences in membrane properties between malignant and non-malignant cells. This remains a preclinical hypothesis.

  • Cytotoxic activity against a range of cancer cell lines in vitro
  • Tumor-growth inhibition reported in some xenograft studies
  • Proposed selectivity for cancer over normal cells requires further validation

Side Effects & Tolerability

Because human studies are lacking, tolerability is unknown; observations are limited to laboratory models. Any use is confined to research settings.

Store lyophilized peptide cold and desiccated, protected from light and moisture. Reconstituted aliquots should be refrigerated or frozen with minimal freeze-thaw cycles.
