Semaglutide

Overview

Semaglutide is a synthetic analog of the incretin hormone glucagon-like peptide-1 (GLP-1), approved in multiple countries for type 2 diabetes and, at a higher dose, for chronic weight management. A fatty-acid side chain that binds albumin extends its half-life, supporting once-weekly subcutaneous (and once-daily oral) dosing. It is among the most extensively studied peptides in metabolic medicine.

Mechanism

Semaglutide is a potent and selective agonist of the GLP-1 receptor. Receptor activation enhances glucose-dependent insulin secretion, suppresses glucagon release, slows gastric emptying, and reduces appetite through central pathways, together producing glycemic control and weight loss.

Dosing

Approved use is by subcutaneous injection once weekly, with an oral formulation also available; doses are titrated over several weeks to limit gastrointestinal side effects. Research-peptide contexts mirror GLP-1 pharmacology but are not clinically validated.
