Overview
Sermorelin is a synthetic peptide corresponding to the first 29 amino acids of human growth-hormone-releasing hormone (GHRH 1-29), the minimal fragment with full secretagogue activity. It stimulates the pituitary to release growth hormone in a pulsatile manner. It has been used in diagnostic growth-hormone stimulation testing and appears in research and clinic protocols exploring support of the endogenous GH/IGF-1 axis.
Mechanism
Sermorelin binds the GHRH receptor on pituitary somatotrophs, increasing growth-hormone synthesis and release, which in turn raises circulating IGF-1. Because it acts upstream of growth hormone itself, its effects depend on intact pituitary function and remain subject to normal somatostatin negative feedback, distinguishing it from direct growth-hormone administration.
Dosing
Research and clinic protocols typically use daily subcutaneous injection, often in the evening, while diagnostic testing uses an intravenous bolus with serial GH sampling. Regimens are protocol-specific and are not an approved therapeutic standard in most countries.
